RAPD+ISSR fingerprinting approach has been undertaken aimed at differentiation of highbush blueberry (Vaccinium corymbosum) cultivars. Genotyping certificates on the basis of RAPD+ISSR analysis has been developed for seven released in Belarus cultivars of V. corymbosum, which can serve as standards for the certification of planting material and collections of the culture in vitro. Using the instrument Bioanalyzer-2100 allows standardizing the acquisition and processing of results, increasing the resolution and comparability of data, which is important when carrying out large-scale certification of samples of highbush blueberry.

Heat shock-dependent changes of catalase activity in wild type Arabidopsis thaliana and in Apx2 knock-out mutant

Changes in catalase (CAT) activity upon heat shock were evaluated in wild type plants of Arabidopsis thaliana (ecotype Col 0) and in ascorbate peroxidase 2 (Apx2)knock-out mutant KO-25. In wild type A. thaliana, increase of CAT activity was found upon moderate heat stress (37°C) and in post-stress recovery phase. In contrast, the effects were less pronounced or absent in KO-25. The induction of catalase activity was more evident upon severe heat shock (44°C). After 2 hours treatment CAT activity was, respectively, 3,0 and 2,3 times higher in wild type and mutant plants comparing to the control probes incubated at room temperature. In post-stress recovery phase at room temperature CAT activity remained increased in wild type, but dropped to the control level in mutant plants. Taking together our data show that CAT appears to be involved in heat shock response in A. thaliana. However, the loss of APX2 activity in the mutant KO-25 is not compensated by additional activation of CAT. Instead, a reduced generation of hydrogen peroxide in the mutant plants can be supposed.

Expression manifestation of mass storage roots and sugar content in top-cross MS hybrids developed to involve lines-pollinators to be separated out from the groups of donors for starting populations, Y752 and KM2, that distinguished with various combinations of high and low absolute values of traits, was considered. Coincidence between empiric and theoretical value distribution as well as correlation relationship between them was found. Lines showing combining ability by productivity and sugar content were identified, their frequency of occurrence in the original forms of Uladovska selection was studied.

Genetic peculiarities on nineteen allozyme loci of fifty-four trees Picea abies (L.) Karst. which differed in periodicity of "fructification" during ten-year-old supervision in introduction plantation of Donbass have been investigated. In the allocated groups of trees, which formed a yield of cones in one, two, three, four and five different years of supervision, differences in genetic variability were revealed. For trees with high reproductive abilities heterozygosity level was close to an average on plantation, while the least – for trees with the lowered reproduction.

The investigation of U. pictorum European area genogeographic structure was carried out using our own and literature data on allozymic variation. South- and North-European population groups, characterized with tendencies to alternative allele fixation were established. The situation corresponds to the model of two allospecies connected with comparatively wide zone of gene introgressions. The southern allospecies, isolated from northern one with the Carpathians and the Alps, is supposed to have its origin from the South-European refugium, whereas the North-European one is the result of recolonization from the eastern, most probably, South-Urals refugium.

The expression of bacterial gene cel7, coding ?-1,4- glucanase in differentiated, de- and redifferentiated tobacco tissues has been studied. Different-level expression of the cel7 transgene was revealed in line pe4: the repression of the cel7 gene in the leaf tissue, inducing the expression in callus and its retention in the adventitious shoots, derived from callus cells was found. Differing expression of bacterial ?-1,4-glucanase in the tobacco leaf tissues affected the phytohormone status of these tissues and as a result reflected in their callusogenic activity and morphogenic potential. It was revealed that under the influence of the recessive ap1-1, bp-1 and tfl1-2 genes A. thaliana instead of a typical tassel there were formed the inflorescences with a new combination of traits atypical to the Brassicaceae family. It was found that the type of Arabidopsis inflorescence can be controlled by individual genes as well as complementary gene interaction where each gene is characterized by its specific effect.

In the genome of Polyommatus icarus two variants of 5S rDNA repeats were identified (Lepidoptera: Lycaenidae). These variants differ considerably by length and sequence of the intergenic spacer region (IGS) although individual copies of rDNA repeats of the same structural variant are nearly identical as a result of concerted evolution. The IGS is composed of microsatellite–related repeats that appeared at the early stages of Lepidoptera radiation. Numerous base substitutions and indels were accumulated in the IGS during subsequent evolution.

Winter wheat breeding for disease resistance started at the V.M. Remeslo Myronivka Institute of Wheat as far back as before the Second World War, but it has become large-scale since 1978. Against the artificial infectious backgrounds of pathogens there were identified the effective sources of resistance both to individual diseases and group of them, studied their donor properties. Based on them new genotypes with group resistance to diseases, tolerant to lodging, showing high productivity and winter-hardiness as well as good grain qualities were generated. Based on disease resistant lines cultivars as follows were developed: Myronivs’ka napivintensyvna, Myronivs’ka 33, Monolog, Ekonomka.

The main genetic methods necessary for the early diagnostic and prognosis of the origin of hereditary determined neoplasia of large bowel was demonstrated in this work. It was noted the significance of genealogic analysis as the first step in the examining of probands for the determining of the contribution of the hereditary factors in the origin of these diseases. It was confirmed the necessity of the complex of contemporary methods of genodiagnostic and karyotype analysis of neoplasia for the introduction in Ukraine. These genetic methods of diagnostics should be used in solution of the important medical and social problem of modern society – early diagnostics of the hereditary determined neoplasia. It will allow decreasing the volume of operative intervention and mortality of these diseases.
